Abstract

Sunset Yellow FCF was tested for 28-days in male Hsd:SD® rats for its potential effect on sperm quality parameters at dietary concentrations of 6,000, 12,000 and 18,000 ppm, corresponding to target doses of 500, 1000, and 1500 mg/kg bw/day. The measured average daily intake was 490, 944, and 1,475 mg/kg bw/day, based on feed consumption and stability of Sunset Yellow FCF in the diet. The animals fed diets with Sunset Yellow FCF presented no clinical signs of toxicity and no differences in feed consumption, body weights, organ weights, ophthalmology, hematology, clinical chemistry, urinalysis, or coagulation parameters that were considered adverse. No mortality or abnormalities were observed at necropsy, and no microscopic changes were observed in histopathology. Increased testes weights relative to body weight in animals of the middle and high intake groups were not associated with any abnormal findings in histopathology. Sperm quality evaluation presented no adverse effects on sperm motility, epididymal sperm count, homogenization-resistant spermatid count, or sperm morphological development. Therefore, in the absence of any adverse effects under the conditions of this study, the NOAEL for Sunset Yellow FCF was 1,475 mg/kg bw/day in male rats, corresponding to 18,000 ppm in the diet.

Abstract

A novel adsorbent consisting of polyethyleneimine-modified multi-wall carbon nanotubes (PEI-MWCNTs) was synthesized by grafting PEI on the carboxyl MWCNTs. The surface properties of the PEI-MWCNTs were measured by scanning electron microscopy, thermogravimetric analysis, Fourier transform infrared, and zeta potential. The adsorption behavior of the PEI-MWCNTs was investigated using sunset yellow FCF as adsorbate. The effects of dosage of adsorbent, the initial pH of solution, contact time and temperature on the adsorption capacity were studied. Then, the kinetics and thermodynamics of the adsorption process were further investigated. Experimental results showed that the adsorption kinetics fitted a pseudo-second-order model and the adsorption isotherms agreed well with the Langmuir model. The adsorption process occurred very fast and the adsorption capacity of PEI-MWCNTs was much higher than that of many of the previously reported adsorbents. Additionally, the plausible adsorption mechanism was discussed.

Abstract

The assembly of small molecules into larger structures, often driven by noncovalent interactions such as hydrogen bonding, aromatic stacking interactions, and burial of hydrophobic surface, is of widespread interest. The interaction of small molecules with aggregates also has a large range of applications from fluorescence aggregation assays to gas storage in framework materials. Here, we utilize nuclear magnetic resonance spectroscopy to investigate the interaction of a small-molecule probe on the assembly state of sunset yellow across a wide range of relative concentrations. Information from both macroscopic (diffusion) and microscopic (chemical shifts) measurements allows the interaction to be studied and the binding mode to be interrogated. Using fluorophenol as the small-molecule probe, we show that the aggregation behavior of sunset yellow is broadly unaffected by the relative amount of fluorophenol added.
